Okay, my 2000 f150, not sure what trans it has... When I'm driving, in town or highway, the truck shifts hard and the tranny squeals. Any idea what the issue could be?

Has the tranny been serviced within the last 50,000 mi? If not the first thing I'd try is a filter and fluid change.

I serviced it when I bought it last year and I'm coming up on 50000, so I'll do it again. What if that doesn't fix it?

Before u change the fluid make sure it's a darker color if it's not don't waist your time, shifting hard could be a speed sensor, or if it only happens when your putting stress on the trans ( going up hit, pulling, increasing speed, or pressing the gas a good deal amount) it could be your torque converter slipping. I had a similar issue, surprising enough the day before I planned on taking it to ford i changed the coil packs and it stopped? So it's hard to tell.

Does it do it under hard throttle or just cruising?

If its doing what mine did, it made like a thunking sound. In fourth gear when the tq converter it locked. And unfortunately mines doing it again
