A misfire is usually an issue with the spark plug/spark plug wire not delivering a large enough spark to fully ignite/burn the fuel/air mixture in the cylinder. It can also be caused by the timing being off but that would usually cause all cylinders to misfire and the engine to run very rough. So basically once you find the misfiring cylinder replace the plug and if need be the wire as well.
Easy way to tell is to run it an pull one wire off at a time to see witch one doesn't make a difference in the way engine is running and if you find one definatly the one!
